目的制备莪术油亚微乳,并考察乳化剂的HLB值对亚微乳理化性质的影响。方法使用高压微射流纳米分散仪制备含不同HLB值乳化剂的莪术油亚微乳;激光粒度及电泳分析仪测定其粒径和表面电位;离心法测定其稳定常数;测定药物在不同HLB值乳化剂的水相中的溶解度,并采用透析法测定亚微乳的体外释放。结果制备得到的莪术油亚微乳粒径在123~154nm之间,粒子带负电。几种乳化剂对药物均有明显的增溶作用,增溶倍数从5.5~8.2倍不等,载药亚微乳的体外释放符合Higuchi方程,且能持续释放48h。结论乳化剂的HLB值不同,制备得到的亚微乳的性质如粒径和稳定性有所不同;随着HLB值增加,乳化剂对药物的增溶作用增强,药物从亚微乳中释放略有加快。
Objective To prepare Curcuma oil submicroemulsion and investigate the effect of emulsifier HLB value on the physicochemical properties of submicroemulsion. Methods High-pressure micro-jet nanodispersion instrument was used to prepare the essential oil of Curcuma zedoana with different HLB emulsifier. The particle size and surface potential were measured by laser particle size and electrophoresis analyzer. The stability constant was determined by centrifugation. Agent in aqueous phase and the dialysis method was used to determine the in vitro release of submicron emulsion. Results The zeaxanthin submicroemulsion particle diameter was between 123 and 154 nm, and the particles were negatively charged. Several emulsifiers have obvious solubilization effect on the drug, solubilization multiples ranging from 5.5 to 8.2 times, in vitro release of drug-loaded submicrobial emulsion in line with Higuchi equation, and sustained release of 48h. Conclusion The HLB values of the emulsifiers are different, and the properties of the prepared submicroemulsions are different, such as particle size and stability. With the increase of the HLB value, the solubilizing effect of the emulsifier on the drug is enhanced and the drug is released from the microemulsion Have to speed up.
