研究细胞因子 (肿瘤坏死因子TNF α )、细胞间质成分 (透明质酸HA )与细胞粘附分子 (CD4 4 )在肾病综合征中的关系及作用。经尾静脉一次性注射阿霉素构建肾病综合征大鼠模型 ,注射阿霉素 14d ,大鼠肾病综合征模型建立。采用放射免疫法及流式细胞术检测TNF α、HA及外周血单核细胞CD4 4阳性细胞数。结果显示模型组血清及肾组织TNF α、HA浓度明显高于对照组 (P <0 0 1)。CD4 4阳性率显著增高 ,与对照组比较有显著性差异 (P <0 0 1) ,肾病综合征CD4 4表达上调与HA浓度增高呈正相关 ,HA与TNF α亦呈正相关。CD4 4与TNF α、HA共同参与了肾病综合征的发病过程。
To study the relationship and role of cytokines (tumor necrosis factor TNFα), interstitial component (hyaluronic acid HA) and cell adhesion molecule (CD4 4) in nephrotic syndrome. The rat model of nephrotic syndrome was established by injecting doxorubicin through the tail vein once a day for 14 days and the nephrotic syndrome model was established. Radioimmunoassay and flow cytometry were used to detect the number of CD4 (4) positive cells of TNFα, HA and peripheral blood mononuclear cells. The results showed that the concentrations of TNFα and HA in serum and kidney of the model group were significantly higher than those of the control group (P <0.01). The positive rate of CD4 4 was significantly higher than that of the control group (P <0.01). The upregulation of CD4 4 in nephrotic syndrome was positively correlated with the increase of HA concentration. HA and TNF α also showed a positive correlation. CD4 4 and TNFα, HA are involved in the pathogenesis of nephrotic syndrome.
