前文曾报道在波长350 nm处测定制剂中硝苯啶(nifedipine,NIF)的含量。但当NIF的浓度小于20μg/ml时,不能进行直接准确的测定。考虑到NIF在波长237 nm处的吸收峰远高于350 nm(相差约4.7倍),对于样本中含量较低的NIF的分析,可以测定样本溶液在237 nm照射前后的吸收值之差(△A_(237))得以解决。本文对此进行了实验,并应用于离体透皮吸收的研究。
It has been reported previously that the content of nifedipine (NIF) in a preparation is measured at a wavelength of 350 nm. However, when the concentration of NIF is less than 20 μg / ml, direct and accurate determination can not be performed. Taking into account that the absorption peak of NIF at wavelength of 237 nm is much higher than 350 nm (a difference of about 4.7 times), for the analysis of the lower content of NIF in sample, the difference between the absorption values of sample solution before and after irradiation at 237 nm A_ (237)) is resolved. This article has carried on the experiment, and applies to the transdermal absorption in vitro research.
