目的建立板蓝根中fructopyrano-(1→4)-glucopyranose(FG)的高效液相色谱-蒸发光散射器含量测定方法。方法色谱柱为Agilent XDB-C18(4.6 mm×150 mm,5μm),流动相为甲醇-水(5∶95),漂移管温度为115℃,载气流速为3.0 L.min-1。结果在5 min内可完成对FG的测定,FG在8.40~21.00 ng内,其对数值与峰面积对数值线性关系良好(r=0.999 0),加样回收率为97.27%~102.72%。结论此方法准确、可靠、快速、灵敏、重现性好,可用于板蓝根中FG的含量测定。
Objective To establish a method for the determination of fructopyrano- (1 → 4) -glucopyranose (FG) in Radix isatidis by high performance liquid chromatography with evaporative light diffuser. Methods The column was Agilent XDB-C18 (4.6 mm × 150 mm, 5 μm). The mobile phase consisted of methanol-water (5:95), the drift tube temperature was 115 ℃ and the carrier gas flow was 3.0 L.min-1. Results The determination of FG was completed within 5 min. The logarithm of the logarithm of peak area was linear (r = 0.999 0) with the range of 8.40-21.00 ng. The recoveries were 97.27% -102.72%. Conclusion The method is accurate, reliable, rapid, sensitive and reproducible. It can be used for the determination of FG in Radix Isatidis.
