目的:牙周病是由多种因素引起的,特别是人牙周膜细胞的缺失。转化生长因子-β1(TGF-β1)是一种多功能细胞因子,在治疗牙周病中发挥重要的作用,但很少有人清楚地研究TGF-β1对人牙周膜细胞的影响。因此,本研究的目的是探讨TGF-β1诱导人牙周膜细胞细胞骨架重排的信号通路。方法:人牙周膜细胞取自健康的前磨牙,并向同步化处理的细胞中加入10 ng/ml的TGF-β1,并通过相差显微镜观察它们的形态学变化。通过免疫组化和共聚焦显微镜观察F-肌动蛋白重排。用Western blot分析蛋白表达情况。结果:我们发现TGF-β1诱导人牙周膜细胞细胞骨架重排,激活ROCK蛋白的表达,并增加p-lIMK和p-cofilin的蛋白表达。ROCK抑制剂Y-27632使ROCK,p-lIMK和p-cofilin的蛋白表达下降。结论:TGF-β1可以诱导人牙周膜细胞细胞骨架重排,并且是通过上凋ROCK,p-lIMK和p-cofilin的活性完成的。本研究可以增强对TGF-β1在治疗牙周疾病方面的作用机制的了解。
Purpose: Periodontal disease is caused by many factors, especially the loss of human periodontal ligament cells. Transforming growth factor-β1 (TGF-β1) is a multifunctional cytokine that plays an important role in the treatment of periodontal disease. However, the effect of TGF-β1 on human periodontal ligament cells has rarely been studied clearly. Therefore, the purpose of this study was to investigate the signaling pathway of TGF-β1-induced cytoskeletal rearrangement in human periodontal ligament cells. METHODS: Human periodontal ligament cells were obtained from healthy premolar and 10 ng / ml TGF-β1 was added to the synchronized cells and their morphological changes were observed by phase contrast microscopy. F-actin rearrangements were observed by immunohistochemistry and confocal microscopy. Western blot analysis of protein expression. RESULTS: We found that TGF-β1 induced cytoskeletal rearrangements in human periodontal ligament cells, activated the expression of ROCK protein and increased the protein expression of p-lIMK and p-cofilin. The ROCK inhibitor Y-27632 decreased the protein expression of ROCK, p-lIMK and p-cofilin. CONCLUSION: TGF-β1 induces cytoskeletal rearrangements of human periodontal ligament cells and is mediated by up-regulation of the activity of ROCK, p-IIMK and p-cofilin. This study may enhance the understanding of the mechanism of action of TGF-β1 in the treatment of periodontal disease.
