目的:探讨槲皮素对人鼻咽癌CNE2细胞生长与凋亡的作用。方法:应用台盼蓝拒染法检测20,40,60,80μmol·L-1浓度槲皮素作用细胞24,48,72 h后的生长状况,流式细胞仪检测在以上浓度下作用细胞48 h后的细胞凋亡率,Western blot法检测以上浓度槲皮素作用细胞48 h后Bcl-2蛋白的表达情况。结果:各浓度槲皮素对人鼻咽癌CNE2细胞的生长有显著的抑制作用,呈明显的时间和剂量依赖性;经不同浓度的槲皮素作用48 h后,细胞凋亡率显著增加,当浓度在40μmol·L-1以上时,与对照组相比,差异具有统计学意义(P<0.01);Western blot法检测Bcl-2蛋白的表达随着槲皮素浓度的增加而降低,槲皮素浓度在40μmol·L-1以上时,与对照组相比,差异具有统计学意义(P<0.01)。结论:槲皮素可以有效抑制人鼻咽癌CNE2细胞生长,诱导细胞凋亡,其诱导该细胞凋亡的作用机制可能是通过下调Bcl-2蛋白的表达。
Objective: To investigate the effect of quercetin on the growth and apoptosis of human nasopharyngeal carcinoma CNE2 cells. Methods: The growth of quercetin-treated cells at 20, 40, 60 and 80μmol·L-1 concentrations for 24, 48, and 72 h were detected by trypan blue exclusion. Flow cytometry h after the apoptosis rate, Western blot method to detect the above concentration of quercetin-induced cells after 48 h Bcl-2 protein expression. Results: Quercetin at various concentrations significantly inhibited the growth of human nasopharyngeal carcinoma CNE2 cells in a dose- and time-dependent manner. After 48 h of quercetin treatment, the apoptosis rate was significantly increased, When the concentration was above 40μmol·L-1, the difference was statistically significant (P <0.01). Western blot showed that the expression of Bcl-2 protein decreased with the increase of quercetin concentration, Compared with the control group, the difference was statistically significant (P <0.01) when the concentration of dermatin was above 40μmol·L-1. CONCLUSION: Quercetin can effectively inhibit the growth of human nasopharyngeal carcinoma CNE2 cells and induce apoptosis. The mechanism of apoptosis induced by quercetin may be through down-regulating the expression of Bcl-2 protein.
