目的 研究金丝桃苷 (hyperin ,Hyp)对大鼠心肌缺血再灌注诱导的细胞凋亡的保护作用。方法 用荧光显微镜和电子显微镜观察 ;原位末段标记法 (TUNEL)和流式细胞术检测等。结果 Hyp(2 5 ,5 0mg·kg- 1 )对缺血 30min后再灌注 3 5h导致大鼠心肌细胞凋亡有保护作用 ;对培养的乳大鼠心肌细胞缺氧 6h再给氧 2 4h引起的细胞凋亡 ,Hyp(0 5~ 5 0 0 μmol·L- 1 )可以减少凋亡细胞的形成。结论 金丝桃苷对缺血再灌注引起的大鼠心肌细胞损伤有保护作用 ;其作用机制可能与金丝桃苷抗心肌细胞凋亡有关。
Objective To study the protective effect of hyperin (Hyp) on apoptosis induced by myocardial ischemia-reperfusion in rats. Methods Fluorescence microscopy and electron microscopy; TUNEL and flow cytometry were performed. Results Hyp (25, 50 mg · kg -1) had a protective effect on the cardiomyocyte apoptosis induced by ischemia 30 min and reperfusion 35 h. The cultured cardiomyocytes were exposed to oxygen for 24 h and reoxygenated for 24 h Hyp (0 5 ~ 500 μmol·L -1) can reduce the formation of apoptotic cells. Conclusions Hyperin has a protective effect on myocardial cell injury induced by ischemia-reperfusion in rats. The mechanism may be related to the anti-myocardial cell apoptosis of hyperoside.
