目的:探讨抑癌基因WTX在结肠癌组织中的表达水平及其意义。方法:收集经病理学证实的50例结肠癌患者的癌组织及其癌旁组织与远癌组织标本,应用荧光定量PCR(qPCR)及Western blot法检测各组织WTX基因及蛋白的表达水平。结果:相对于远癌组织(1.00±0.00),癌组织与癌旁组织WTX基因表达量分别为0.36±0.17和0.91±0.34,各组织间差异均有统计学意义(均P<0.05)。WTX蛋白的相对表达量在癌组织,癌旁组织,远癌组织分别为0.27±0.11,0.64±0.23,0.72±0.19,各组织间差异均有统计学意义(P<0.05)。结论:抑癌基因WTX的表达降低可能在结肠癌的发生发展进程中起重要作用。
Objective: To investigate the expression of tumor suppressor gene WTX in colon cancer and its significance. Methods: Fifty cases of colon cancer tissues and their adjacent tissues and distant cancer tissues were collected by pathology. The expression of WTX gene and protein in each tissue was detected by qPCR and Western blot. Results: The WTX gene expression was 0.36 ± 0.17 and 0.91 ± 0.34 in cancer tissues and adjacent non-cancerous tissues (1.00 ± 0.00), respectively, with significant difference (all P <0.05). The relative expression of WTX protein was 0.27 ± 0.11,0.64 ± 0.23 and 0.72 ± 0.19 in cancer tissues, paracancerous tissues and distant cancer tissues, respectively. There was significant difference among all the tissues (P <0.05). Conclusion: The decreased expression of tumor suppressor gene WTX may play an important role in the development of colon cancer.
