本文报道一种用薄层层析法分析~(55)Fe标记的分枝菌素作分枝杆菌种间鉴别的方法,敏感性强、快速简便,有可能用于临床常规检查。本法用11种分枝杆菌,培养在含适量甲磺酸去铁胺、联吡啶或环糊精的低铁琼脂斜面,收集菌落以甲醇或酒精抽提分枝菌素,用0.02uci~(55)FeCl标记,将标记标本滴在二氧化硅凝胶60薄层层析板上,在石油精:丁醇:乙酸乙酯2:3:3溶剂中层析分离,用放射分析系统(AMBIS)检测放射活性斑点。结果表明,生长快的耻垢分枝杆菌、偶遇分枝杆菌NCTC 10394和M.neoaurum在37℃
This paper reports a thin layer chromatography method for the analysis of ~ (55) Fe-labeled mycobacteria for identification of mycobacterial species. It is sensitive, rapid and simple, and may be used for clinical routine examination. This law with 11 kinds of mycobacteria cultured with appropriate amount of deferoxamine mesylate, bipyridine or cyclodextrin low-iron agar slants collected colony methanol or alcohol mycobacterium extract with 0.02uci ~ ( 55) FeCl. The labeled sample was dropped onto a silica gel 60 TLC plate and chromatographed on a petroleum spirit: butanol: ethyl acetate 2: 3: 3 solvent system and analyzed by a radiometric analysis system (AMBIS ) Detection of radioactive spots. The results show that fast growing Mycobacterium smegmatis, occasionally encountered in Mycobacterium NCTC 10394 and M. neoaurum at 37 ℃
