本文用大鼠从体内、外观察了前列腺素E_2(PGE_2)对肝再生的作用。重点集中在肝部分切除后所分离的离体枯否细胞(KC)在大肠杆菌脂多糖(LPS)的刺激下合成及分泌PGE_2的能力。结果显示,给大鼠体内注射消炎痛(5mg/kg)抑制KC的PGE_2产生,从而显著地阻碍了肝脏的再生。其DNA合成率明显低于未使用组(P<0.01)。而在DNA合成高峰期(术后24h)所分离的再生肝之KC,其合成PGE_2的能力明显强于假手术组对等数量的KC(PGE_2浓度分别为15ng/ml及6.5ng/ml,P<0.01)。据此我们得出结论,KC通过其内源性PGE_2的产生从而启动肝细胞的再生。
The purpose of this study was to investigate the effect of prostaglandin E2 (PGE2) on liver regeneration in vivo and in vitro. The focus is on the ability of ex vivo Kupffer cells (KCs) isolated after partial hepatectomy to synthesize and secrete PGE 2 stimulated by E. coli lipopolysaccharide (LPS). The results showed that intraperitoneal injection of indomethacin (5 mg / kg) in rats inhibited PGE 2 production by KC, thereby significantly hindering liver regeneration. The DNA synthesis rate was significantly lower than the unused group (P <0.01). However, the KC of regenerated liver isolated during the peak period of DNA synthesis (24h after operation) was significantly stronger than that of the sham-operated group (PGE 2 concentrations of 15 ng / ml and 6.5 ng / ml, P <0.01). From this we conclude that KC initiates the regeneration of hepatocytes through its production of endogenous PGE2.
