目的:观察表达嵌合体蛋白PAcA/CTB转基因番茄防龋疫苗的免疫原性和免疫反应性。方法:新西兰大白兔随机分为实验组、阴性对照组和阳性对照组3组,分别喂食3个剂量(目的蛋白量相差1.5倍)的转基因番茄汁、非转基因番茄汁和变异链球菌灭活全菌疫苗。每周免疫1次,连续免疫4周,ELISA法检测免疫前和免疫后不同时期血液、唾液样品中抗变异链球菌PAc的IgG、IgA抗体效价。结果:口服免疫后实验组与阳性对照组大白兔血液和唾液中均出现特异性的抗PAc抗体,抗体水平可维持数周,而阴性对照组抗体水平变化不明显。结论:表达嵌合体蛋白PAcA/CTB的转基因番茄能够诱导动物机体产生有效的系统免疫应答和黏膜免疫应答。
OBJECTIVE: To observe the immunogenicity and immunoreactivity of PAcA / CTB transgenic tomato dental caries vaccine expressing chimeric protein. Methods: New Zealand white rabbits were randomly divided into experimental group, negative control group and positive control group. Three groups of transgenic tomato juice, non-transgenic tomato juice and streptococcus mutans were inactivated by 3 doses (target protein difference 1.5 times) Bacterial vaccine. Immunization was performed once a week for 4 weeks. The titer of IgG and IgA antibodies against Streptococcus mutans PAc in blood and saliva before and after immunization were detected by ELISA. Results: Anti-PAc antibodies were found in the blood and saliva of rabbits in the experimental group and the positive control group after oral immunization for several weeks, while the antibody levels in the negative control group did not change significantly. Conclusion: The transgenic tomato expressing the chimeric protein PAcA / CTB can induce an effective systemic immune response and mucosal immune response in the animal body.
