目的:观察益水生新饮对12周龄自发性高血压大鼠(SHR)肾脏组织中TGF-β1、CTGF、FN信号通路的影响,揭示益水生新饮对自发性高血压大鼠肾脏的保护及其作用靶点。方法:将50只12周龄雄性SHR大鼠随机分为:益水生新饮低剂量组[35.55 g/(kg·d)简称低剂量组]、益水生新饮中剂量组[71.10g/(kg·d)简称中剂量组]、益水生新饮高剂量组[142.20g/(kg·d)简称高剂量组]、苯那普利组[10mg/(kg·d)]、模型组,每组10只,模型组生理盐水灌胃,每天1次,共8周。8周后处死大鼠,检测各组大鼠肾组织中TGF—β1、CTGF、FN mRNA和蛋白的检测。观察各组大鼠肾脏病理改变和血压改变。结果:(1)与模型组相比,益水生新饮组和苯那普利组血压明显降低,差异有显著性(P<0.01),其中高剂量组与苯那普利组相比差异有显著性(P<0.01);(2)病理检查提示,与益水生新饮组和苯那普利组相比,模型组系膜细胞及基质增生病变明显加快。(3)与模型组相比,益水生新饮组及苯那普利组肾脏组织中TGF-β1、CTGF及FN表达均下调,差异有显著性(P<0.05,P<0.01),其中,益水生新饮高剂量组与苯那普利组相比,CTGF下调的差异有显著性(P<0.01)。结论:益水生新饮主要通过抑制肾脏中TGF-β1、CTGF及FN信号通路的表达,从而减轻SHR大鼠早期肾脏肾小球硬化和肾间质纤维化的程度,起到延缓高血压的进展和肾脏保护的作用。
Objective: To observe the effects of YISHUI Sheng Xin Yin on TGF-β1, CTGF and FN signaling pathways in kidney of spontaneously hypertensive rats (SHR) at 12 weeks of age and to reveal the protective effects of YISHUI Sheng Xin Yin on the kidneys of spontaneously hypertensive rats And its target. (2) Pathological examination showed that compared with Yishuanshengxin drink group and benazepril group, the mesangial cells and matrix hyperplasia in model group were significantly accelerated. (3) Compared with the model group, the expression of TGF-β1, CTGF and FN in Yishuanshengxin drink group and benazepril group were all significantly decreased (P <0.05, P <0.01) Yisui Sheng Xin Yin high dose group and benazepril group, CTGF down the difference was significant (P <0.01). Conclusion: Yishui Sheng Xin Yin can retard the progression of hypertension by inhibiting the expression of TGF-β1, CTGF and FN in kidney, and thus reducing the degree of early renal glomerulosclerosis and interstitial fibrosis in SHR rats And the role of kidney protection.
