应用Sanger的双脱氧链终止法,对本室构建的人类白细胞介素2基因重组子pRET-90的440bpcDNA功能片段(NcoI克隆点的HgiAI-DraI片段)进行DNA碱基序列分析。结果表明:白细胞介素2cDNA功能片段按照正确的方向和翻译顺序连接在载体pET3d的NcoI-BamHI切点之间。
The nucleotide sequence of the 440bp cDNA fragment (HgiAI-DraI fragment of NcoI cloning site) of human interleukin 2 gene recombinant pRET-90 constructed by Sanger was analyzed by DNA sequencing using Sanger’s dideoxy chain termination method. The results showed that the functional fragment of interleukin 2 cDNA was ligated between the NcoI-BamHI cleavage sites of the vector pET3d in the correct orientation and translation order.
