阳离子脂质简化了核酸序列转入真核细胞的过程,最初用于转染的脂质包括L_(IPOFECTIN)及L_(IPOFECT) ACE~(TM)、DOTAP,它们都是单价阳离子。L_(IPOFECT) AMINE,一种新的脂质体配方包含一种带有精胺头部的多聚阳离子脂质,可以在大多数类型的细胞中提高转染效率,比单价阳离子脂质体试剂的活性高出30倍。本报道通过转染和人原代成纤维细胞将L_(IPOFECT) AMINE与其它几种可购买到的单阳离子脂质试剂进行了比较。同时对转染条件的确定和优化进行了阐述。
Cationic lipids simplify the transfer of nucleic acid sequences into eukaryotic cells. Lipids originally used for transfection include IPOFECTIN and IPOFECT ACE ™, DOTAP, which are all monovalent cations. L IPOFECT AMINE, a new liposomal formulation containing a polycationic lipid with spermine head, can enhance transfection efficiency in most types of cells compared to monovalent cationic liposome agents 30 times more active. This paper compared L IPOFECT AMINE with several other commercially available mono-cationic lipid agents by transfection and human primary fibroblasts. At the same time, the conditions of transfection were determined and optimized.
