目的:探讨苯丙素苷类抗氧化剂异毛蕊花苷对人胃癌MGC 803细胞生长和分化的影响.方法:以1.5%二甲基亚砜为阳性对照,采用细胞计数、肿瘤形成率实验、酶活性测试、流式细胞仪分析和West-ern blot等方法分别评价异毛蕊花苷对MGC 803细胞生长、致瘤性、碱性磷酸酶(ALP)及乳酸脱氢酶(LDH)活性、细胞周期和周期相关基因表达的影响.结果:MGC 803细胞经异毛蕊花苷处理后,细胞生长和增殖呈浓度和时间依赖性抑制;异毛蕊花苷20μmol/L可使细胞致瘤性明显受抑,胃癌细胞分化标志酶ALP和 LDH活性均呈时间依赖性下降,细胞周期表现为G_0/G_1期阻滞,G_1→S期调控点相关蛋白p53、p21/WAF1和p16/INK4表达上调,C-myc蛋白表达受抑.结论:异毛蕊花苷可明显抑制MGC 803细胞生长及逆转胃癌细胞恶性表型进而诱导分化,这可能与细胞周期G_0/G_1期阻滞及其对周期相关基因表达的调控作用有关.
Objective: To investigate the effect of isatin on the growth and differentiation of human gastric cancer cell line MGC 803.Methods: Using 1.5% dimethylsulfoxide (DMSO) as positive control, the cell counts, tumor formation rate and enzyme activity Test, flow cytometry analysis and West-ern blot were used to evaluate the effect of isatin on the growth, tumorigenicity, alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) activity, cell cycle and cycle Related gene expression.Results: MGC 803 cells were treated with isocitosine, the cell growth and proliferation was inhibited in a concentration-dependent and time-dependent manner; Isoverbasin 20 μmol / L significantly inhibited cell tumorigenicity, differentiation of gastric cancer cells The activities of ALP and LDH decreased in a time-dependent manner, and the cell cycle showed G_0 / G1 arrest. The expressions of p53, p21 / WAF1 and p16 / INK4 in G_1 → S phase were up-regulated while the expression of C-myc was down-regulated .Conclusion: Isoverbasicin can significantly inhibit the growth of MGC 803 cells and reverse the malignant phenotype of gastric cancer cells, which may be related to the arrest of cell cycle G_0 / G_1 and the regulation of cell cycle related gene expression.
