目的观察血管紧张素转化酶抑制剂洛汀新对阿霉素肾病(AN)大鼠肾脏血管紧张素Ⅱ(ATⅡ)浓度及受体表达的影响。方法采用逆转录-聚合酶链反应(RT-PCR)技术检测大鼠肾组织血管紧张素Ⅱ-Ⅰ型受体(AT1R)mRNA表达情况,以放射免疫分析法测定血浆及肾组织ATⅡ含量。结果洛汀新可降低AN大鼠血浆及肾组织ATⅡ含量,使肾脏AT1RmRNA表达增强。结论洛汀新对AN大鼠肾脏AT1RmRNA表达的影响与降低血浆及肾组织ATⅡ含量有关。
Objective To investigate the effect of angiotensin converting enzyme inhibitor Lotensin on renal angiotensin Ⅱ (AT Ⅱ) concentration and its receptor expression in adriamycin nephropathy rats. Methods The expression of angiotensin Ⅱ type Ⅰ receptor (AT1R) mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR) in rat kidney. AT Ⅱ contents in plasma and kidney tissue were determined by radioimmunoassay. Results Lotensin can reduce AT Ⅱ content in plasma and kidney of AN rats, and enhance the expression of AT1R mRNA in kidney. Conclusion The effect of Lotensin on the expression of AT1R mRNA in the kidney of AN rats is related to the decrease of AT Ⅱ in plasma and kidney.
