目的:建立适合甲巯咪唑血药浓度快速测定的HPLC法。方法:色谱柱:Hypersil ODS柱(150 mm×4.6 mm,5μm);流动相:甲醇-0.05%乙酸溶液(1:99);波长:252 nm。结果:该方法在含甲巯咪唑血浆浓度0.1~2.0μg/ml及2.0~50.0μg/ml范围内定量线性良好(r分别为0.9994和0.9999),最低检测限为1ng。精密度和回收率均良好。结论:本方法适用于服用甲巯咪唑及卡比马唑的临床病人的血药浓度测定,以便进行临床药学研究。
Objective: To establish a HPLC method suitable for rapid determination of methimazole plasma concentration. Methods: Column: Hypersil ODS column (150 mm × 4.6 mm, 5 μm); mobile phase: methanol-0.05% acetic acid solution (1:99); wavelength: 252 nm. Results: The method was linear in the range of 0.1 ~ 2.0μg / ml and 2.0 ~ 50.0μg / ml with methimazole (r = 0.9994 and 0.9999, respectively). The detection limit was 1ng. Precision and recovery are good. Conclusion: This method is suitable for the determination of plasma concentrations in clinical patients taking methimazole and carbimazole for clinical pharmacy research.
