目的:评价小槐花Desmodium caudatum(Thunb.)DC.提取物对α-葡萄糖苷酶的抑制活性。方法:通过建立体外α-葡萄糖苷酶抑制模型,对小槐花提取物进行活性筛选。结果:小槐花提取物均有较好的α-葡萄糖酶抑制作用,其中以乙酸乙酯部位(DCEA)和石油醚部位(DCPE)活性较好(IC50=9.79,39.2 mg·L-1),其次为正丁醇部位(DCBU)(IC50=625.1 mg·L-1),抑制活性均远远大于阳性对照阿卡波糖(IC50=1 103.01 mg·L-1)。DCBU对α-葡萄糖苷酶抑制活性呈显浓度依赖性;DCPE和DCEA对α-葡萄糖苷酶抑制活性对浓度的变化较灵敏,随浓度的增加而急剧上升。结论:小槐花石油醚部位和乙酸乙酯部位对α-葡萄糖苷酶活性抑制非常显著,具有开发治疗糖尿病降血糖药物的价值。
Objective: To evaluate the inhibitory activity of extract of Desmodium caudatum (Thunb.) DC. Against α-glucosidase. Methods: The inhibitory model of α-glucosidase in vitro was established and the extracts of Sophora japonica L. were screened. Results: All the extracts of Sophora japonica L. had better α-glucosidase inhibitory activity, of which ethyl acetate (DCEA) and petroleum ether (DCPE) had better activity (IC50 = 9.79, 39.2 mg · L -1) , Followed by n-butanol (DCBU) (IC50 = 625.1 mg · L-1). The inhibitory activities were much higher than that of the positive control acarbose (IC50 = 1 103.01 mg · L -1). DCBU showed a concentration-dependent inhibition of α-glucosidase activity; DCPE and DCEA were more sensitive to changes in α-glucosidase inhibitory activity, with the concentration increased sharply. CONCLUSION: The results showed that the inhibition of α-glucosidase activity by petroleum ether fraction and ethyl acetate in Xiaohuai flower is very significant, which has the value of developing drugs for the treatment of hypoglycemic diabetes.
