突触素(P38)是一种位于突触囊泡(Ⅰ、Ⅱ型)膜上的钙连结糖蛋白,在神经递质的释放过程中起着重要作用.突触素的分布准确地反映了突触前终末的分布,同时它也是突触生长的一个标志.关于突触素的年龄相关变化,目前研究很少.实验用C_(57)BL雄性小鼠共31只,分为青年组(5月龄)、成年组(13月龄)和老年组(24月龄).用抗突触素的小鼠单克隆抗体(SY_(38))(ABC法)标记突触素,结合IBAS图象分析系统测定海马CA_1、CA_3区分子层,齿状回分子层中带和小脑I、V叶分子层突触素免疫反应产物光密度(OD值).结果显示,突触素免疫反应产物在海马结构和小脑皮质呈明显的板层分布,被检各区突触素免疫反应产物的密度差异很大(P<0.05).在三个年龄组,被检各区突触素免疫反应产物OD值以青年组最大,成年组次之,老年组最小,各组间差异显著(P<0.05或0.01).在年龄相关的变化中,以小鼠皮质分子层突触素免疫反应产物OD值的下降更显著,和青年组比较,对照组Ⅰ、Ⅱ在海马结构平均降低20.39%、33.29%,在小脑皮质平均降低29.38%、52.64%.结果表明,突触素在小脑海马结构和小脑皮质的分布具有区域性,随着年龄的增加,突触素含量逐渐减少.
