用离体大鼠心脏冠状动脉前降支结扎后再灌流模型观察了硫酸镁对再灌流心律失常的预防作用。第一批实验显示再灌流前缺血时间与室颤发生率呈钟形曲线关系。第二批实验在冠状动脉结扎前1min将硫酸镁浓度升至3.6,4.8和6.0mmol/L,室颤总发生率呈剂量依赖性降低,分别由对照组的100%(1.2mmol MgSO_4)降至82%,73%和18%(P<0.01)。再灌流前硫酸镁升至6.0mmol/L则无抗心律失常作用。缺血前硫酸镁升至6.0mmol/L的同时将钙升高1.5mmol/L,硫酸镁的抗心律失常作用消失。结果表明硫酸镁具有一定的抗再灌流心律失常作用,该作用可能是由直接或间接的钙拮抗作用所致。
The preventive effect of magnesium sulfate on reperfusion arrhythmia was observed by the reperfusion model of isolated rat cardiac anterior descending coronary artery. The first experiments showed a bell-shaped curve of the ischemic time before reperfusion and the incidence of ventricular fibrillation. The second experiment increased the concentration of magnesium sulfate to 3.6, 4.8 and 6.0 mmol / L 1 min before coronary artery ligation and the total incidence of ventricular fibrillation in a dose-dependent manner decreased from 100% (1.2 mmol MgSO 4) in the control group 82%, 73% and 18% (P <0.01). Magnesium sulfate rose to 6.0mmol / L before reperfusion without anti-arrhythmic effect. Pre-ischemic magnesium sulfate rose to 6.0mmol / L while calcium increased 1.5mmol / L, magnesium sulfate anti-arrhythmic effect disappeared. The results show that magnesium sulfate has a certain anti-reperfusion arrhythmia, the effect may be caused by direct or indirect calcium antagonism.
