目的:评价新型抗体靶向性核酸转移系统及其核心组份[葡萄球菌A蛋白-聚乙烯亚胺交联物(SPAPEI交联物)]对培养细胞的毒性作用。方法:将SPA-PEI交联物与转铁蛋白受体的抗体和绿色荧光蛋白质粒以适宜的质量比混合,组装成一种抗体靶向性核酸转移系统;采用不同浓度SPA-PEI交联物或抗体靶向系统分别处理Hep G2人肝癌细胞不同时间后MTT比色法测定细胞的存活率。结果:SPA-PEI交联物对培养的Hep G2人肝癌毒性作用有明显的浓度依赖性和时间依赖性,但其杀伤效应较PEI降低;抗体靶向系统的细胞毒作用较弱,其作用细胞的存活曲线近似于SPA。结论:SPA-PEI交联物对培养细胞有较强的杀伤效应,但以其为核心组份构建的抗体靶向性核酸转移系统无明显细胞毒性作用。
OBJECTIVE: To evaluate the cytotoxicity of novel antibody targeting nucleic acid transfer system and its core component [Staphylococcal protein A-polyethyleneimine (SPAPEI)] on cultured cells. Methods: The SPA-PEI conjugate and transferrin receptor antibody and green fluorescent protein plasmid were mixed with the appropriate mass ratio, assembled into an antibody targeting nucleic acid transfer system; using different concentrations of SPA-PEI cross-linked material or Antibody targeting system were used to determine the survival rate of Hep G2 human hepatocellular carcinoma cells by MTT colorimetric assay after different time. Results: The conjugate of SPA-PEI had significant concentration-dependent and time-dependent effects on cultured Hep G2 human hepatocarcinoma cells, but its killing effect was lower than that of PEI. The cytotoxicity of SPA-PEI conjugate system was weak, The survival curve approximates SPA. CONCLUSION: The conjugate of SPA-PEI has strong killing effect on cultured cells, but the antibody-targeted nucleic acid transfer system constructed with it as the core component has no obvious cytotoxicity.
