目的:观察大蒜素对糖尿病大鼠心肌细胞凋亡及MAPK信号传导通路的影响。方法:应用链脲佐菌素建立糖尿病大鼠模型,随机分为糖尿病模型组、大蒜素低、中和高剂量组,同时设正常对照组。大蒜素各剂量组按不同浓度比例喂饲大蒜素。连续干预16天后处死剪取左心室组织,TUNEL检测心肌细胞凋亡情况,Western blotting检测MEK、ERK和P38表达情况。结果:模型组心肌细胞凋亡程度较重,大剂量组心肌细胞凋亡情况改善明显;模型组MEK、ERK表达下降,P38表达增加,大剂量组的表达改善明显。结论:大蒜素可有效防治糖尿病心肌细胞凋亡,激活MEK-ERK通路以保护心肌,同时降低P38蛋白表达。
Objective: To observe the effect of allicin on cardiomyocyte apoptosis and MAPK signal transduction pathway in diabetic rats. Methods: Diabetic rats were induced by streptozotocin and randomly divided into diabetic model group, allicin low dose group, middle dose group and high dose group. At the same time, normal control group was established. Allicin dose groups according to different concentrations of allicin feeding. The left ventricular tissues were sacrificed after 16 days of continuous intervention. Cardiomyocyte apoptosis was detected by TUNEL. The expressions of MEK, ERK and P38 were detected by Western blotting. Results: The cardiomyocyte apoptosis in the model group was more serious, and the apoptosis in the high dose group was improved obviously. The expression of MEK and ERK in the model group decreased, the expression of P38 increased and the expression in the high dose group improved obviously. Conclusion: Allicin can effectively prevent and treat diabetic cardiomyocyte apoptosis, activate MEK-ERK pathway to protect myocardium and reduce P38 protein expression.
