本试验对小鼠脚掌皮下接种癌细胞后引起的,以淋巴结为主的淋巴网状组织的反应,进行了组织学及超微结构的检查。癌细胞接种后,无论局部还是非局部,淋巴结反应均很显著。早期以副皮质区体积扩大、免疫母细胞数目增加为主要特点;局部和非局部淋巴结免疫母细胞数目分别在接种后第4天和第7天达到高峰,并以局部增加最为明显。晚期生发中心扩大融合,髓索浆细胞数目增加。中期为早、晚期变化的过渡和移行阶段。提示淋巴结针对肿瘤的免疫反应早期可能是以细胞免疫为主,晚期可能以体液免疫为主,中期细胞免疫继续存在,同时体液免疫开始增强。局部淋巴结在参与肿瘤免疫方面反应早,而且比较强烈,非局部淋巴结在体液免疫方面可能发挥更大作用。透射电镜观察发现,淋巴结内窦组织细胞可以直接吞噬癌细胞,该过程可能需要淋巴细胞辅助。实验结果提示副皮质区免疫母细胞数目增多可作为抗肿瘤细胞免疫的指标,对于带瘤宿主预后判断有重要意义。
In this study, we examined the histological and ultrastructural responses of lymph node-based lymphoid reticulum caused by subcutaneous inoculation of mouse paw. After inoculation of cancer cells, both local and non-local lymph node reaction were significant. In the early stage, the volume of the paracortex enlarged and the number of immunoblasts increased. The number of immunoblasts in both local and non-local lymph nodes peaked on the 4th and 7th day after inoculation, and the most significant increase was local. The late germinal center expands and merges, the pulp marrow cell number increases. Mid-early and late changes in the transition and migration phase. Prompt lymph node tumor response to the early may be based on cellular immunity, late may be the main humoral immunity, mid-term cellular immunity continue to exist, while humoral immunity began to increase. Local lymph nodes respond earlier and more strongly to tumor immunity, and nonlocal lymph nodes may play a greater role in humoral immunity. Transmission electron microscopy revealed that the lymph nodes within the sinus tissue cells can directly devour cancer cells, the process may require lymphocyte-assisted. The experimental results suggest that the increase of the number of immunoblastic cells in the paracortex may be used as an indicator of anti-tumor cell immunity, which is of great significance for the prognosis of tumor-bearing hosts.
