目的:研究神经元谷氨酸(Glu)合成、分解和分泌与癫痫发病的关系。方法:用致病剂马桑内酯(CL)处理培养大鼠海马神经元,用高效液相色谱仪、液体闪烁仪分别检测培养神经元内外Glu含量和神经元内谷氨酸脱羧酶(GAD)活性。结果:与对照组比较,CL致痫神经元内Glu浓度没有显著差异,神经元外Glu浓度显著升高(P<0.05),神经元内GAD活性显著增高(P<0.05)。结论:癫病发病中,痫性神经元Glu合成、分解、分泌增强。
Objective: To study the relationship between neuronal glutamate (Glu) synthesis, decomposition and secretion and epilepsy. Methods: Hippocampal neurons were cultured with the causalitide (CL), and the levels of Glu and neuronal glutamate decarboxylase (GAD) were measured by HPLC and liquid scintillation respectively )active. Results: Compared with the control group, there was no significant difference in Glu concentration in neurons of CL-induced epilepsy. Glu concentration in neurons increased significantly (P <0.05) and GAD activity in neurons increased significantly (P <0.05). Conclusion: During the onset of epilepsy, the synthesis, decomposition and secretion of Glu in epileptic neurons are enhanced.
