目的探讨PTEN基因转染对人舌癌细胞系SCC-4凋亡的影响及其作用机制。方法将转染PTEN的SCC-4(pEGFP-PTEN-SCC-4)细胞作为实验组,以SCC-4和pEGFP-SCC-4作为对照,流式细胞仪检测3组细胞凋亡情况;应用Western blotting法检测3组中Akt、p-Akt、Bim的表达情况。结果流式细胞仪检测结果显示,实验组细胞凋亡率比对照组明显增高(P<0.01);Western blotting结果显示,各组细胞间总Akt水平差异不明显;p-Akt在实验组中表达明显低于对照组;Bim在实验组中表达明显高于对照组。结论 PTEN转染能够诱导SCC-4的凋亡,可能机制是负调控PI3K/Akt信号通路并上调Bim的表达。
Objective To investigate the effect of PTEN gene transfection on the apoptosis of human tongue cancer cell line SCC-4 and its mechanism. Methods The SCC-4 (pEGFP-PTEN-SCC-4) cells transfected with PTEN were used as experimental group and the apoptosis of three groups was detected by flow cytometry with SCC-4 and pEGFP-SCC-4 as controls. The expressions of Akt, p-Akt and Bim in three groups were detected by blotting. Results The results of flow cytometry showed that the apoptotic rate in the experimental group was significantly higher than that in the control group (P <0.01). The results of Western blotting showed that there was no significant difference in the total Akt between the groups. The expression of p-Akt in the experimental group Significantly lower than the control group; Bim in the experimental group was significantly higher than the control group. Conclusions PTEN transfection can induce the apoptosis of SCC-4. The possible mechanism is that it negatively regulates the PI3K / Akt signaling pathway and up-regulates Bim expression.
