建立了葛根中岩藻糖、鼠李糖、阿拉伯糖、半乳糖、葡萄糖、甘露糖、木糖、核糖、甘露醇、木糖醇、果糖等11种单糖的离子色谱方法,并应用此方法分析了不同产地葛根多糖中单糖组成及含量。葛根多糖经超声提取,Box-Behnken响应面优化超声提取条件,三氟乙酸水解,以Carbo Pac PA20(150 mm×3 mm)色谱柱分离,20 mmol/L NaOH溶液等度洗脱。结果表明,11种单糖的定量限为0.03~0.07 mg/kg,在0.2~5 mg/L线性范围内线性关系良好(r~2>0.9990),加标回收率为80.3%~112.4%。经Box-Behnken响应面优化后的最佳提取工艺为液料比28:1(mL/g),超声时间44 min,超声温度65℃。经离子色谱方法测定不同产地的葛根样品中均含有岩藻糖、鼠李糖、葡萄糖,其中葡萄糖的含量最高含量在114.3~622.2 mg/g之间。方法可为葛根质量评价提供技术支撑。
An ion chromatography method was established for the determination of eleven monosaccharides, such as fucose, rhamnose, arabinose, galactose, glucose, mannose, xylose, ribose, mannitol, xylitol and fructose in Radix Puerariae. The composition and content of monosaccharide in Pueraria lobata from different areas were analyzed. Pueraria lobata polysaccharides were extracted by ultrasonic, Box-Behnken response surface was optimized by ultrasonic extraction, trifluoroacetic acid hydrolysis, Carbo Pac PA20 (150 mm × 3 mm) column, 20 mmol / L NaOH solution isocratic elution. The results showed that the limit of quantification of 11 kinds of monosaccharides was 0.03-0.07 mg / kg, the linearity was good (r ~ 2> 0.9990) in the linear range of 0.2-5 mg / L, and the recoveries were 80.3% -112.4%. The optimized extraction process of Box-Behnken response surface was 28: 1 (mL / g), ultrasonic time was 44 min and ultrasonic temperature was 65 ℃. The kudzu root samples from different areas were determined by ion chromatography, which contained fucose, rhamnose and glucose. The highest content of glucose was 114.3 ~ 622.2 mg / g. The method can provide technical support for the quality evaluation of Radix Puerariae.
