目的 :研究转化生长因子 βI型受体 (TβRⅠ )与胆囊癌细胞增殖的关系 ,并探讨TβRⅠ在胆囊癌变过程中的作用机理。方法 :应用S P免疫组织化学技术对 35例原发性胆囊癌 ,1 0例胆囊腺瘤及 1 0例慢性胆囊炎进行TβRⅠ及增殖细胞核抗原 (PCNA)两种基因蛋白的检测。 结果 :胆囊癌中TβRⅠ阳性率为 34 .2 9% ,显著低于胆囊腺瘤、胆囊炎 (阳性率均为 70 % ) (P <0 .0 5) ,伴淋巴结和远处转移者TβRⅠ阳性表达率明显低于无转移者 (P <0 .0 5) ,Nevin分期与TβRⅠ强阳性表达率呈负相关 (r =- 0 .3986 ,P =0 .0 1 77)。胆囊癌TβRⅠ表达水平与增殖细胞核抗原标记指数 (PCNALI)呈负相关 (r =- 0 .40 2 4 ,P =0 .0 1 66)。TβRⅠ表达与病理组织学分级及性别、年龄无关。结论 :胆囊癌TβRⅠ表达减少 ,可促进肿瘤细胞的增殖、浸润和转移 ;TβRⅠ的低表达而导致的转化生长因子 β(TGFβ)负性生长调控作用的逃逸 ,可能是胆囊癌变过程中的重要机制
Objective: To investigate the relationship between transforming growth factor β receptor (TβRⅠ) and proliferation of gallbladder carcinoma cells and to explore the mechanism of TβRⅠ in gallbladder carcinogenesis. Methods: S P immunohistochemistry was used to detect TβRⅠ and PCNA in 35 cases of primary gallbladder carcinoma, 10 cases of gallbladder adenoma and 10 cases of chronic cholecystitis. Results: The positive rate of TβRI in gallbladder carcinoma was 34.29%, which was significantly lower than that of gallbladder adenoma and cholecystitis (both positive rate was 70%) (P <0.05), and the positive rate of TβRI in lymph nodes and distant metastasis The expression rate of Nevin was negatively correlated with the strong positive rate of TβRⅠ (r = - 0.3986, P = 0.0117). The expression of TβRI in gallbladder carcinoma was negatively correlated with the PCNA index (r = - 0.40 2 4, P = 0.0166). TβR Ⅰ expression and histopathological grading and sex, age has nothing to do. Conclusion: The decrease of TβRI expression in gallbladder carcinoma can promote the proliferation, invasion and metastasis of tumor cells. The escape of the negative growth regulatory factor of transforming growth factor β (TGFβ) induced by the low expression of TβRⅠ may be an important mechanism in carcinogenesis of gallbladder
