目的:探讨SD大鼠慢性非细菌性前列腺炎(CAP)炎症组与正常对照组前列腺平滑肌细胞(PSMCs)内游离钙离子浓度([Ca2+]i)的差异及其在CAP中的意义。方法:利用SD大鼠前列腺蛋白提取液与弗氏完全佐剂(FCA)诱导出CAP模型,体外贴壁培养并纯化炎症组与正常对照组PSMCs,细胞经钙离子荧光探针FLUO-3AM孵育后在激光共聚焦显微镜(LSCM)下连续动态扫描。结果:CAP组与对照组的PSMCs钙离子荧光强度分别为80.39±9.00和27.95±10.04,差异具有显著性(P<0.01)。结论:CAP SD大鼠PSMCs[Ca2+]i明显升高,其升高可能增强了PSMCs的收缩作用,从而引起CAP的一系列如对疼痛敏感性增强、下腹部疼痛不适的症状。
Objective: To investigate the difference of intracellular free calcium concentration ([Ca2 +] i) in prostate smooth muscle cells (PSMCs) of chronic non-bacterial prostatitis (CAP) inflammation group and normal control group and its significance in CAP. Methods: CAP model was induced by using SD rat prostatic protein extract and Freund’s complete adjuvant (FCA). PSMCs in inflammatory group and normal control group were cultured and purified in vitro. After the cells were incubated with calcium ion fluorescence probe FLUO-3AM Continuous dynamic scanning under confocal laser scanning microscopy (LSCM). Results: The calcium fluorescence intensity of PSMCs in CAP group and control group were 80.39 ± 9.00 and 27.95 ± 10.04, respectively, with significant difference (P <0.01). CONCLUSIONS: The [Ca2 +] i of PSMCs in CAP SD rats is significantly increased, which may increase the contractile effect of PSMCs, resulting in a series of symptoms such as increased pain sensitivity and lower abdominal pain discomfort.
