目的 检测谷胱甘肽S转移酶(GST-π)、P-糖蛋白在骨肉瘤、骨软骨瘤、正常骨组织中的表达及其与骨肉瘤临床病理改变之间的关系。 方法 对53例原发性骨肉瘤、15 例骨软骨瘤、17 例正常骨组织进行免疫组化染色。 结果 在骨肉瘤中GST-π、P-糖蛋白的表达明显高于正常骨组织和骨软骨瘤,骨肉瘤中P-糖蛋白的表达与术前有无化疗明显相关,在骨肉瘤、骨软骨瘤和正常骨组织中P-糖蛋白与GST-π共同表达率分别为52.8% , 13.3% 和0。 结论 (1)骨肉瘤患者化疗不敏感可能与肿瘤组织中自发性存在P-糖蛋白和GST-π过表达有关;(2)术前化疗能明显提高P-糖蛋白的表达,提示骨肉瘤患者预后差可能与自发性和获得性P-糖蛋白的过表达有关;(3)联合检测GST-π、P-糖蛋白可作为鉴别良恶性骨肿瘤的有益指标。
Objective To detect the expression of glutathione S transferase (GST-π) and P-glycoprotein in osteosarcoma, osteochondroma and normal bone tissue and their relationship with clinical pathological changes of osteosarcoma. Methods Immunohistochemical staining was performed on 53 cases of primary osteosarcoma, 15 cases of osteochondroma, and 17 cases of normal bone tissue. Results The expression of GST-π and P-glycoprotein in osteosarcoma was significantly higher than that in normal bone tissue and osteochondroma. The expression of P-glycoprotein in osteosarcoma was significantly related to the preoperative chemotherapy. In osteosarcoma and osteochondral The co-expression rates of P-glycoprotein and GST-π in tumor and normal bone tissues were 52.8%, 13.3% and 0, respectively. Conclusion (1) Insensitivity of chemotherapy to osteosarcoma may be related to the spontaneous expression of P-glycoprotein and GST-π in tumor tissue; (2) Preoperative chemotherapy can significantly increase the expression of P-glycoprotein, suggesting that osteosarcoma patients The poor prognosis may be related to the overexpression of spontaneous and acquired P-glycoprotein; (3) The combined detection of GST-π, P-glycoprotein can be used as a useful indicator to identify benign and malignant bone tumors.
