为进一步为钩体基因工程疫苗的研究提供理论和实验依据,采用纯化的强毒017钩体39kd疏水外膜蛋白(OmpL39)疫苗免疫豚鼠,对其免疫力进行检定并观察该疫苗对神经体液和红细胞免疫功能的影响。结果显示:OmpL39在豚鼠体内能产生高效价的阳性抗体(MAT>1:3,200),免疫力合格;在免疫应答期,OmpL39疫苗对豚鼠外周血5羟色胺(5HT)、去甲肾上腺素(NE)、多巴胺(DA)等有明显影响,其中5HT、DA、NE明显降低,5羟吲哚乙酸明显升高,与空白对照组有显著性差异(P<005);OmpL39对红细胞免疫功能也有明显影响,其中红细胞cαb受体粘附酵母菌花环率,红细胞免疫复合物花环率均明显升高,红细胞免疫粘附因子促进率上升,抑制率下降,两者比值显著增大(P<005)。提示:在免疫应答期OmpL39除了增强体液免疫外,还能刺激机体影响神经递质的释放,对神经体液免疫功能有明显的调节作用,同时可提高红细胞免疫功能
In order to provide theoretic and experimental basis for the further study on the genetic engineering vaccine of Leptospira, the guinea pigs were immunized with the purified 39kd hydrophobic OmpL39 vaccine and its immunity was observed. Impact of Erythrocyte Immune Function. The results showed that: OmpL39 in guinea pigs can produce high titer of positive antibodies (MAT> 1: 3,200), immunity is qualified; in the immune response period, OmpL39 vaccine on guinea pig peripheral blood 5-HT (5 HT) Epinephrine (NE), dopamine (DA) and other significant effects, including 5 HT, DA, NE was significantly lower, 5 hydroxyindoleacetic acid was significantly increased, and the control group were significantly different (P <0 05 ); OmpL39 also had a significant effect on the immune function of erythrocytes, in which erythrocyte cαb receptor adhesion rate of yeast rosette, erythrocyte immune complex rosette were significantly increased, erythrocyte immune adherent factor increased, the inhibition rate decreased, the ratio between the two Significantly increased (P <0 05). Tip: In the immune response period OmpL39 in addition to enhancing humoral immunity, but also stimulate the body to affect the release of neurotransmitters, neuroendocrine immune function significantly regulate the role, while improving the immune function of red blood cells
