机械应力是一种普遍存在的刺激因子。我们以前的工作表明,机械应力能调节人红细胞膜上葡萄糖运输蛋白-GluT-1的功能:GluT-1介导的输入、输出葡萄糖的速率能对机械应力产生响应[1]。但到底这种功能变化的原因是来源于机械应力对GluT-1的结构改变还是其它因子至今尚不清楚。为此,我们试图从GluT-1内源性色氨酸的荧光变化上观测机械应力对GluT-1结构的影响,并探讨其可能的机制。
Mechanical stress is a ubiquitous stimulus. Our previous work showed that mechanical stress regulates the function of glucose transporter-GluT-1 on the human erythrocyte membrane: the rate of GluT-1 -mediated import and export of glucose can respond to mechanical stress [1]. But in the end this functional change is due to mechanical stress on GluT-1 structural changes or other factors is still not clear. To this end, we try to observe the effect of mechanical stress on the GluT-1 structure from the change of fluorescence of GluT-1 endogenous tryptophan and to explore its possible mechanism.
