目的 探讨单克隆抗体ND1对大肠癌的诊断价值。方法 利用放射免疫法对荷人结肠癌裸鼠模型进行显像研究。结果 99mTc直接标记抗人结肠癌单克隆抗体ND1及F(ab’) 2 片段 ,标记率为 94 %和 87%。标记后抗体的免疫活性良好。注入荷人结肠癌裸鼠体内 16小时后 ,肿瘤组织中出现较高的特异性浓聚 ,肿瘤轮廓显示清晰 ,F(ab’) 2 片段的放射本底消除快 ,图像质量优于ND1。体内分布显示 :ND1及F(ab’) 2 片段可与移植瘤特异性结合 ,肿瘤与正常器官组织的放射比值 (T/NT)、F(ab’) 2 片段显著高于ND1,ND1在肿瘤、血液及器官组织均有较高的放射性摄取 ,不利于放射性本底的消除。结论 99mTc F(ab’) 2 对大肠癌的放射免疫显像图像清晰 ,优于99mTc ND1,为其临床应用提供了实验依据
Objective To investigate the diagnostic value of monoclonal antibody ND1 in colorectal cancer. Methods Radioimmunoassay was used to study the colorectal cancer xenograft model in nude mice. Results 99mTc directly labeled anti-human colon cancer monoclonal antibodies ND1 and F(ab’) 2 fragments, and the labeling rate was 94% and 87%. The labeled antibody has good immune activity. After 16 hours of inoculation into nude mice bearing human colon cancer, a high specific concentration of tumor was observed in the tumor tissue, the tumor outline was clearly shown, and the radiation background of the F(ab’)2 fragment was eliminated quickly, and the image quality was superior to that of ND1. The in vivo distribution showed that ND1 and F(ab’)2 fragments could specifically bind to xenografts. The ratios of tumor (T/NT) and F(ab’)2 fragments were significantly higher than those of ND1 in tumors and normal organ tissues. , blood and organ tissues have high radioactivity, which is not conducive to the elimination of radioactive background. Conclusion 99mTc F(ab’) 2 has clear radioimmunoimages for colorectal cancer and is superior to 99mTc ND1, providing experimental basis for its clinical application.
