目的:总结转录因子Oct-4在人类肿瘤组织中的表达情况及其与肿瘤生物学行为、肿瘤放化疗抵抗等之间的关系。方法:应用PubMed数据库检索系统,以“Oct-4、肿瘤和干细胞”为关键词,检索1996-2008年关于Oct-4研究的文献,共检索到英文文献476篇。纳入标准:1)Oct-4的功能及调控;2)Oct-4在肿瘤中的表达;3)肿瘤组织Oct-4基因表达的临床意义。根据标准,纳入分析22篇参考文献。结果:转录因子Oct-4属POU家族蛋白,在动物早期胚胎发育过程中起着重要作用,其通过激活或抑制下游靶基因来参与维持细胞的多向潜能性及未分化状态。已在多种实体肿瘤细胞中发现Oct-4异常高表达,由此更进一步证实了肿瘤干细胞学说。Oct-4的表达可能与肿瘤的恶性程度及生物学行为、肿瘤放化疗抵抗密切相关。结论:有关Oct-4的研究可能为判断肿瘤预后提供新的指标,也有利于发展新的抗癌药物作用靶点。
OBJECTIVE: To summarize the expression of transcription factor Oct-4 in human tumor tissue and its relationship with the biological behavior of tumor and the resistance of tumor to radiotherapy and chemotherapy. Methods: The PubMed database search system was used to search the literature of Oct-4 from 1996 to 2008 with the keywords of “Oct-4, tumor and stem cell”. A total of 476 English articles were retrieved. Inclusion criteria: 1) the function and regulation of Oct-4; 2) the expression of Oct-4 in tumor; 3) the clinical significance of Oct-4 gene expression in tumor tissue. According to the standard, included in the analysis of 22 references. Results: The transcription factor Oct-4 belongs to the POU family of proteins and plays an important role in early embryonic development of animals. It participates in maintaining the multi-directional potential and undifferentiated state of cells by activating or inhibiting downstream target genes. Oct-4 abnormally high expression has been found in a variety of solid tumor cells, thus further confirming the theory of tumor stem cells. The expression of Oct-4 may be closely related to the malignancy and biological behavior of the tumor and the chemoresistance of the tumor. Conclusions: The study of Oct-4 may provide new indicators for judging the prognosis of the tumor and also be helpful for the development of new anti-cancer drug targets.
