Objective: To establish a simple, stable and high success rate of New Zealand rabbit myocardial cell isolation method. Methods: The effects of perfusion rate of Langen-dorff perfusion device [5 mmol / min (8 ml / min) and 65 mmol / min (11 ml / min)] on cell isolation were compared during cell dissociation. Results: Compared with the perfusion rate of 50 r / min, the time required for perfusion was 65 r / min [(33 ± 9.10) min vs (19.29 ± 2.73) min vs 50% vs 91% , (38.26 ± 2.63)% (85.71 ± 2.21)%] (P <0.01). CONCLUSION: Perfusion velocity is an important factor affecting the acute myocardial isolation in New Zealand rabbits. The perfusion rate at 65 r / min can be more stable and single cardiomyocytes can be successfully obtained.
