目的:观察黄芪金樱子合剂对氧化低密度脂蛋白(OX-LDL)诱导大鼠肾系膜细胞凋亡的影响及干预途径。方法:应用不同浓度(高、中、低)黄芪金樱子合剂的大鼠含药血清,与OX-LDL(浓度为100μg/ml)及大鼠肾系膜细胞(RMC)共同培养24h,采用流式细胞仪检测细胞凋亡指标Annexin V、Caspase 3和逆转录-聚合酶联扩增法(RT-PCR)观察Caspase 3、Bcl-2/Bax、Fas、FasL表达的变化。结果:不同浓度黄芪金樱子合剂可显著减少OX-LDL诱导的RMC凋亡细胞数目(P<0.05),抑制Caspase 3的基因和蛋白质表达(P<0.05),降低Fas和FasL的基因表达(P<0.05),提高Bcl-2/Bax的比率(P<0.05),但量效关系不明显。结论:黄芪金樱子合剂具有改善OX-LDL诱导大鼠肾系膜细胞凋亡的药理学效应。
OBJECTIVE: To observe the effect and mechanism of Astragalus roxburghii mixture on apoptosis of rat mesangial cells induced by oxidized low-density lipoprotein (OX-LDL). METHODS: Rat serum containing different concentrations (high, middle and low) of Astragalus sculpina mixture was incubated with OX-LDL (100 μg/ml) and rat mesangial cells (RMC) for 24 hours. The expression of caspase 3, Bcl-2/Bax, Fas and FasL were detected by Annexin V, Caspase 3 and reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: Different concentrations of Astragalus Scutellariae Mixture could significantly reduce the number of apoptotic RMC cells induced by OX-LDL (P<0.05), inhibit the expression of Caspase 3 gene and protein (P<0.05), and reduce the expression of Fas and FasL genes ( P<0.05), increased Bcl-2/Bax ratio (P<0.05), but the dose-effect relationship was not significant. Conclusion: Astragalus roxburghii can improve the apoptosis of rat mesangial cells induced by OX-LDL.
