目的研究北极刺参岩藻聚糖硫酸酯(Thelenata anaxsulfated fucan,Ta-FUC)对2型糖尿病小鼠胰岛素抵抗症状的改善作用。方法以高脂高糖饮食建立2型糖尿病小鼠模型,以含有0.04%和0.16%TaFUC的饲料分别连续饲喂小鼠11周,实验结束后,称量白色脂肪组织重量,检测各组小鼠空腹血糖、葡萄糖耐受量,及空腹血清胰岛素、脂联素、瘦素、肿瘤细胞坏死因子-α(TNF-α)含量。结果Ta-FUC可降低模型小鼠体重(P<0.05),减少小鼠白色脂肪组织重量(P<0.01);并显著降低模型小鼠空腹血糖(P<0.05),提高口服葡萄糖耐受量(P<0.01),降低空腹血清胰岛素水平(P<0.05);同时还可降低血清中瘦素和TNF-α的含量(P<0.05,P<0.01)。结论北极刺参岩藻聚糖硫酸酯对模型小鼠的胰岛素抵抗具有显著的改善作用,其作用机理可能与改善肥胖引起的脂肪组织细胞因子的分泌紊乱有关。
Objective To study the effect of Tailed Forsythiasm (AUC) on the insulin resistance in type 2 diabetic mice. Methods The model of type 2 diabetic mice was established by high-fat and high-sugar diet. The mice were fed with 0.04% and 0.16% TaFUC respectively for 11 weeks. After the experiment, the weight of white adipose tissue was measured, Fasting blood glucose, glucose tolerance, and fasting serum insulin, adiponectin, leptin, tumor necrosis factor-alpha (TNF-α) content. Results Ta-FUC could reduce body weight (P <0.05), decrease the weight of white adipose tissue in mice (P <0.01), reduce the fasting blood glucose (P <0.05) and increase the oral glucose tolerance P <0.01), lower fasting serum insulin level (P <0.05), and also lower serum leptin and TNF-α levels (P <0.05, P <0.01). CONCLUSIONS: Fucoidan significantly improves the insulin resistance in model mice, and its mechanism may be related to the improvement of obesity-induced secretion of cytokines in adipose tissue.
