目的:观察中药复方肾消康对糖尿病肾病大鼠肾脏Alox12基因表达的影响,探讨糖尿病肾病(DN)的发生与Alox12基因变化的相关性,筛选中药复方肾消康防治DN的药效作用靶点。方法:采用链脲佐菌素加高脂饮食的方法建立糖尿病大鼠肾脏损伤的模型。运用美国Affymetrix公司的基因表达谱芯片,尤其是通过聚类分析寻找和糖尿病肾病有重要相关性的基因——Alox12基因是其中之一,并进一步采用实时荧光定量RT-PCR法,观察Alox12基因表达的变化。结果:花生四烯酸12-脂加氧酶(Alox12),在糖尿病大鼠肾脏表达上调,肾消康治疗组表达下调。结论:在糖尿病肾损伤状态下,Alox12表达上调,与糖尿病肾脏损伤有重要相关性,而经肾消康治疗后大鼠肾脏组织中Alox12表达下调,可见肾消康对Alox12基因表达具有良性调节作用,其作用机制还有待于进一步研究。
Objective: To observe the effect of Chinese medicine compound Shenxiazang on kidney Alox12 gene expression in diabetic nephropathy rats, explore the relationship between the occurrence of diabetic nephropathy (DN) and the changes of Alox12 gene and screen the pharmacodynamic target of traditional Chinese medicine Shenzheng Kang in preventing and treating DN . Methods: Streptozotocin plus high fat diet was used to establish a model of kidney injury in diabetic rats. Using Affymetrix gene expression profiling chip from the United States, in particular, through cluster analysis to find genes that have important correlation with diabetic nephropathy - Alx12 gene is one of them, and further using real-time fluorescent quantitative RT-PCR method to observe Alox12 gene expression The change. Results: Arachidonic acid 12-lipoxygenase (Alox12) was up-regulated in the kidney of diabetic rats and down-regulated in the Shenkukang treatment group. CONCLUSION: Alox12 is up-regulated in diabetic renal injury and has an important correlation with diabetic nephropathy. Alox12 expression is down-regulated in renal tissues of rats treated with Shenkangjikang, which shows that Shenkang Kang has a good regulatory effect on Alox12 gene expression , Its mechanism needs to be further studied.
