目的:观察与探讨榄香烯逆转肺癌化疗耐药的作用。方法:采用细胞培养及动物实验分别从体外、体内研究榄香烯逆转化疗耐药的作用;采用噻唑蓝(MTT)法检测由含药血清培养的A549肺腺癌细胞的增殖率;通过复制Lewis肺癌小鼠模型,并予榄香烯及多柔比星进行处理,最终测量各组小鼠肿瘤的体积,计算抑瘤率。结果:细胞研究结果显示,榄香烯、多柔比星及榄香烯+多柔比星组的Optical Density光密度值(OD值)均小于模型组(P<0.01),而榄香烯组OD值高于多柔比星组及榄香烯+多柔比星组(P<0.01),多柔比星组OD值高于榄香烯+多柔比星组(P<0.01)。动物研究结果显示:与模型组比较,榄香烯组、多柔比星组及榄香烯+多柔比星组肿瘤体积均明显缩小(P<0.01),而多柔比星组的抑瘤率高于榄香烯组(P<0.01),低于榄香烯+多柔比星组(P<0.01)。结论:榄香烯具有一定的抑瘤作用,且具有逆转化疗药物耐药的作用。
Objective: To observe and explore the effect of elemene on reversing chemoresistance of lung cancer. Methods: Cell culture and animal experiments were used to study the effect of elemene on chemoresistance in vitro and in vivo. The proliferation rate of A549 lung adenocarcinoma cells cultured with drug-containing serum was detected by MTT assay. Lung cancer mice model, and elemene and doxorubicin treatment, the final measurement of tumor volume of mice in each group, calculate the inhibition rate. Results: The results of cell studies showed that the optical density optical density (OD) values of elemene, doxorubicin and elemene + doxorubicin group were less than those of model group (P <0.01) OD values were higher in doxorubicin group and elemene + doxorubicin group (P <0.01). The OD value of doxorubicin group was higher than that in elemene + doxorubicin group (P <0.01). The results of animal studies showed that the tumor volume of elemene group, doxorubicin group, and elemene + doxorubicin group were all significantly reduced (P <0.01) compared with model group, while the anti-tumor The rate was higher than elemene (P <0.01), lower than elemene + doxorubicin (P <0.01). Conclusion: Elemene has a certain anti-tumor effect, and has the effect of reversing the drug resistance of chemotherapy.
