目的探讨和比较甲状旁腺激素1-34、阿仑膦酸钠、辛伐他汀对卵巢切除大鼠骨折愈合的作用。方法 3月龄SD大鼠40只,随机分为5组,每组8只,除假手术组外,其余大鼠接受双侧卵巢切除术,术后3个月建立股骨骨折模型,并于术后分别给予双蒸水(安慰剂组)、甲状旁腺激素1-34,20μg/kg,5d/周(甲状旁腺激素组)、阿仑膦酸钠70μg/(kg·周)(磷酸钠组)、辛伐他汀5mg/(kg·d)(辛伐他汀组),单纯骨折组不给予药物处理。6周后处死大鼠,双能X线法检测大鼠骨折侧股骨中段骨密度,CR摄片及评分评估骨折愈合情况,三点弯曲试验分析骨折部位最大载荷。结果股骨中段骨密度单纯骨折组显著高于其余四组,甲状旁腺激素组显著高于安慰剂组、辛伐他汀组和磷酸钠组(P<0.05);CR摄片单纯骨折组组、甲状旁腺激素组骨折愈合情况优于其余三组;CR评分甲状旁腺激素组与单纯骨折组显著高于其余三组(P<0.05),其余组间差异无统计学意义;生物力学检测各组最大载荷由高到低为单纯骨折组、甲状旁腺激素组显著高于其余三组(P<0.05),其余组间差异无统计学意义。结论在本研究采用的剂量和干预周期下,仅有甲状旁腺激素1-34能显著促进骨质疏松大鼠骨折愈合。
Objective To investigate and compare the effects of parathyroid hormone 1-34, alendronate and simvastatin on fracture healing in ovariectomized rats. Methods Forty three-month-old Sprague-Dawley rats were randomly divided into 5 groups with 8 rats in each group. Except the sham operation group, the other rats received bilateral ovariectomy and the femur fracture model was established 3 months after operation. (Placebo group), parathyroid hormone 1-34, 20μg / kg, 5d / week (parathyroid hormone group), alendronate 70μg / (kg · week) (sodium phosphate Group) and simvastatin 5 mg / (kg · d) (simvastatin group). The simple fracture group was not given drug treatment. Results In the middle femur, the bone mineral density fracture group was significantly higher than the other four groups. The parathyroid hormone group was significantly higher than the placebo group, simvastatin group and sodium phosphate group (P <0.05) Phecate group fracture healing was better than the other three groups; CR score parathyroid hormone group and simple fracture group was significantly higher than the other three groups (P <0.05), the other groups no significant difference; biomechanical testing of each group The maximal load from high to low was simple fractures group, parathyroid hormone group was significantly higher than the other three groups (P <0.05), the other group differences were not statistically significant. Conclusions Only the parathyroid hormone 1-34 can significantly promote fracture healing in osteoporotic rats at the dose and intervention intervals used in this study.
