目的探讨不同温孵育方式对酶联免疫吸附试验(ELISA)检测HCV抗体结果的影响。方法收集同一批号试剂盒的阴、阳性标准品及同一批号的质控品,将质控品平行检测21孔(B行后三孔依次为空白1孔、N对照1孔、P对照1孔)。除温育方式不同外,其余操作均按照试剂盒说明书进行。3种孵育方法分别为电热恒温水浴箱、电子恒温培养箱、MKF-6酶标版快速孵育器中进行温育。结果采用3种温育方式进行ELISA抗-HCV检测结果比较无显著差异。结论采用电子恒温水浴箱、电子恒温培养箱、MKF-6酶标版快速孵育器等对ELISA法检测结果无明显干扰,但MKF-6酶标版快速孵育器在提高实验效率,缩短实验时间方面具有突出优势,值得同界人士参考借鉴。
Objective To investigate the effect of different incubation methods on the detection of HCV antibody by enzyme linked immunosorbent assay (ELISA). Methods The positive and negative standards of the same batch of kits and the control samples of the same batch were collected. The quality control products were tested in parallel for 21 wells (the first three wells followed by the blank one well, the N control one well and the P control one well) . In addition to different ways of incubation, the remaining operations are carried out in accordance with kit instructions. Three kinds of incubation methods were electric heated water bath, electronic incubator, MKF-6 plate fast incubator incubator. Results There was no significant difference in anti-HCV test results between the three incubation methods. Conclusion The results of ELISA assay showed no significant interference with the ELISA incubator using electronic thermostatted water bath, electronic incubator, MKF-6 plate incubator, etc. However, the MKF-6 plate incubator was effective in improving the experimental efficiency and shortening the experimental time Has outstanding advantages, it is worth for reference with the community.
