为阐明不同药物相互作用数学模型分析结果的差异来源,观察了SAHA与三氧化二砷单独或联合用药对体外培养的人肿瘤细胞及正常细胞的杀伤作用。通过三种药物相互作用数学模型——Loewe相加模型、Bliss独立模型和周氏中效模型,分析了联合用药的实验结果并比较了这三种模型计算结果的区别。结果说明,在量效曲线拟合方法相同的前提下,这三种模型得到的结论是一致的;量效曲线拟合方法的差异,而非模型本身,是不同模型分析结论差异的主要来源。对不同的细胞两药联合的结果并不完全一致,在不同的剂量下联合作用的效果有一定差异。
In order to elucidate the different sources of the results of mathematical model analysis of different drug interactions, the killing effects of SAHA and arsenic trioxide alone or in combination on human tumor cells and normal cells cultured in vitro were observed. Three experimental models of drug interactions, the Loewe additive model, the Bliss independent model and the Zhou’s median effective model, were used to analyze the experimental results of the combination therapy and the differences between the calculated results of the three models. The results show that the results of the three models are consistent under the same premise of the fitting method of quantitative and effective curves. The differences of the fitting methods of the quantitative and effective curves, rather than the model itself, are the main sources of the different conclusions of the model analysis. The results of the combination of the two drugs on different cells are not exactly the same, and the effect of the combined effect is different at different doses.
