目的 建立适用于临床的N myc检测方法 ,提供预后信息。方法 应用荧光原位杂交 (FISH)技术检测N myc基因扩增 ,应用RT PCR和免疫组化技术分别检测N myc转录和蛋白水平表达。结果 N mycmRNA水平和N myc蛋白表达水平之间没有严格的平行关系。结论 FISH和免疫组化技术共同运用于N myc基因扩增和蛋白表达的检测 ,不仅能为NB预后和治疗提供更可靠的分子生物学信息 ,而且有助于进一步探讨N myc基因调节机制和N myc在NB临床行为中的作用。
Objective To establish a clinically applicable N myc assay and provide prognostic information. Methods Fluorescence in situ hybridization (FISH) was used to detect the amplification of N myc gene. RT-PCR and immunohistochemistry were used to detect the expression of N myc in transcription and protein levels, respectively. Results There was no strict parallel relationship between N myc mRNA levels and N myc protein expression levels. Conclusion Both FISH and immunohistochemistry techniques are applied to the detection of N myc gene amplification and protein expression, which not only provides more reliable molecular biology information for NB prognosis and treatment, but also helps to further explore the regulatory mechanism of N myc gene and N The role of myc in NB clinical behavior.
