目的探讨乙醛脱氢酶2(ALDH2)对大鼠百草枯中毒性急性肺损伤(ALI)的保护作用。方法 SD大鼠120只随机均分为五组。A组大鼠作为空白对照;B、C、D和E组以百草枯(80mg/kg)一次性灌胃建立百草枯中毒大鼠ALI模型,2h后按1×10-8 mol/L硫辛酸(C组)、大豆苷(D组)和硫辛酸+大豆苷(E组)灌胃;A、B组给予10%NaOH灌胃作对照。在中毒后6、24、48和72h每组分别取6只大鼠,检测血清超氧化物歧化酶(SOD)的活性和丙二醛(MDA)含量,并检测肺组织线粒体中ALDH2的活性。结果与A组相比,B、C、D和E组血清SOD活性均降低(P<0.05),降低幅度依次为E组>D组>B组>C组;B、C、D和E组血清MDA浓度均增高(P<0.05),且B、D、E组高于C组(P<0.05);B、C、D、E组肺组织线粒体中ALDH2活性均增高(P<0.05),增高幅度依次为C组>B组>D组>E组。结论 ALDH2对百草枯中毒性大鼠ALI有保护作用。
Objective To investigate the protective effect of aldehyde dehydrogenase 2 (ALDH2) on paraquat-induced acute lung injury (ALI) in rats. Methods 120 SD rats were randomly divided into five groups. Rats in group A were used as blank control. Rats in groups B, C, D and E were treated with paraquat (80mg / kg) once by intragastric administration to establish paraquat poisoning ALI model. After 2h, the rats were treated with 1 × 10-8 mol / L lipoic acid (Group C), daidzin (group D) and lipoic acid + daidzin (group E). Groups A and B were given gavage with 10% NaOH as control. At 6, 24, 48 and 72 hours after poisoning, 6 rats in each group were taken to measure the activity of superoxide dismutase (SOD) and malondialdehyde (MDA), and the activity of ALDH2 in lung mitochondria was detected. Results Compared with group A, the serum SOD activities in groups B, C, D and E were decreased (P <0.05), and the decreasing ranges were E group> D group> B group> C group; B, C, D and E groups (P <0.05), and the levels of ALDH2 in group B, D and E were higher than that in group C (P <0.05). The activity of ALDH2 in group B, C, D and E were higher than that in group C Increase in order of C group> B group> D group> E group. Conclusion ALDH2 has protective effects on paraquat-induced ALI in rats.
