目的初步探讨仅通过观察行为学改变对锰中毒大鼠模型的判别作用。方法将24只SPF级雄性SD大鼠按体重随机分为4组,每组6只,分别为对照组、低剂量组(15.0mg/kg)、中剂量组(25.0mg/kg)和高剂量组(50.0mg/kg),染锰3个月,并对其进行行为学评分、肌张力改变、平衡木试验、水迷宫实验几项行为学测试,染锰结束后检测其大脑纹状体酪氨酸羟化酶(Tyrosine Hydroxylase,TH)表达含量,并以对照组大鼠平均水平的30.00%为界划分锰中毒组和非锰中毒组,通过多项行为学指标联合建立判别分析函数。结果锰中毒组大鼠行为学评分、肌张力评分、平衡木评分均高于非锰中毒组(P<0.01),TH表达量锰中毒组低于非锰中毒组(P<0.01);Fisher判别法形成判别函数,符合率为91.67%(22/24),误判率为8.33%(2/24);该判别函数对非锰中毒组和锰中毒组的正确判别符合率均高于85.71%。结论通过行为学评分、肌张力改变、平衡木试验几项行为学测试组合对锰中毒性大鼠模型做判断初步可行。
Objective To investigate the discrimination effect of rat model of manganese poisoning by observing behavioral changes only. Methods Twenty-four SPF male Sprague-Dawley rats were randomly divided into 4 groups according to body weight, with 6 rats in each group. They were control group, low dose group (15.0mg / kg), middle dose group (25.0mg / kg) (50.0 mg / kg) for 3 months. The behavioral score, the change of muscle tone, the balance beam test and the water maze test were used for several behavioral tests. The content of tyrosine ammonia The expression of Tyrosine Hydroxylase (TH) in the rats was divided into two groups: control group (30.00%) and control group. Manganese poisoning group and non-manganese poisoning group were divided into two groups. Discriminant analysis function was established by multiple behavioral indexes. Results The scores of behavior, muscle tone and balance beam in manganese poisoning group were significantly higher than those in non-manganese poisoning group (P <0.01), and the levels of TH expression in manganese poisoning group were lower than those in non-manganese poisoning group (P <0.01) The coincidence rate was 91.67% (22/24) and the false positive rate was 8.33% (2/24). The correctness of the discriminant function was 85.71% for non-manganese poisoning group and manganese poisoning group. Conclusions Several behavioral test combinations such as behavioral score, muscle tone change and balance beam test are feasible for judging manganese poisoning rat model.
