目的研究海洋真菌烟曲霉Aspergillus fumigatus YK-7中的生物碱类代谢产物。方法利用多种色谱方法(硅胶柱色谱、ODS柱色谱、Sephadex LH-20柱色谱、高效液相色谱)进行分离纯化,通过理化性质和波谱数据鉴定化合物的结构。采用台盼蓝法和MTT法进行抗肿瘤活性测试。结果从发酵液和菌丝体中分离得到10个生物碱类化合物,分别鉴定为pseurotin A(1)、pseurotin A1(2)、14-norpseurotin A(3)、FD-838(4)、demethoxyfumitremorgin C(5)、9β-hydroxyverruculogen TR-2(6)、6-methoxyspirotryprostatin B(7)、spiro[5H,10H-dipyrrolo[1,2-a:1’,2’-d]pyrazine-2-(3H),2’-[2H]indole]-3’,5,10(1’H)-trione(8)、terezine D(9)和14-hydroxyterezine D(10)。结论化合物3、6、7、9、10为首次从海洋来源烟曲霉中分离得到。化合物1~4具有一定的抗肿瘤活性。
Objective To study the alkaloid metabolites in marine fungal Aspergillus fumigatus YK-7. Methods The compounds were isolated and purified by a variety of chromatographic methods (silica gel column, ODS column, Sephadex LH-20 column and HPLC). The structures of compounds were identified by their physico-chemical properties and spectral data. Antitumor activity was tested by trypan blue method and MTT assay. Results Ten alkaloids were isolated from the fermentation broth and mycelium and identified as pseurotin A (1), pseurotin A1 (2), 14-norpseurotin A (3), FD-838 (4), demethoxyfumitremorgin C (5) 9β-hydroxyverruculogen TR-2 (6), 6-methoxyspirotryprostatin B (7), spiro [5H, 10H-dipyrrolo [1,2-a: ), 2 ’- [2H] indole] -3’, 5,10 (1’H) -trione (8), terezine D (9) and 14-hydroxyterezine D (10). Conclusion Compounds 3, 6, 7, 9 and 10 were isolated from Aspergillus fumigatus for the first time. Compounds 1 to 4 have some anti-tumor activity.
