目的对化合物2-(3E,5E-庚二烯基)-3,6-二羟基-5-(3-甲基-2-丁烯基)苯甲醛(isodihydroau-roglaucin)的抗癌活性进行研究。方法利用细胞毒实验测定isodihydroauroglaucin对人宫颈癌细胞(HeLa)的生长抑制作用;通过DNA含量分析和Annexin V-FITC/PI双染实验检测isodihydroauro-glaucin诱导的HeLa细胞死亡;利用单细胞凝胶电泳方法检测isodihydroauroglaucin对DNA的损伤作用。结果 Isodihydroauroglaucin对HeLa细胞的生长具有抑制作用,其IC50值为170μmol.L-1;DNA含量分析和Annexin V-FITC/PI双染实验表明isodihydroauroglaucin诱导HeLa细胞凋亡;isodi-hydroauroglaucin对DNA有损伤作用。结论 Isodihydroauroglaucin可能通过诱导DNA损伤诱导He-La细胞凋亡。
Aim To investigate the antitumor activity of 2- (3E, 5E-heptadienyl) -3,6-dihydroxy-5- (3-methyl-2-butenyl) benzaldehyde (isodihydroau-roglaucin) . Methods The inhibitory effect of isodihydroauroglaucin on human cervical carcinoma cells (HeLa) was determined by cytotoxicity assay. The death of HeLa cells induced by isodihydroauro-glaucin was detected by DNA content analysis and Annexin V-FITC / PI double staining assay. Single cell gel electrophoresis Methods The effect of isodihydroauroglaucin on DNA damage was tested. The results showed that Isodihydroauroglaucin had an inhibitory effect on the growth of HeLa cells with an IC50 value of 170 μmol·L-1. DNA content analysis and Annexin V-FITC / PI double staining showed that isodihydroauroglaucin induced HeLa cell apoptosis. . Conclusion Isodihydroauroglaucin may induce HeLa cell apoptosis by inducing DNA damage.
