[目的]观察恩度、紫杉醇对Her-2过表达乳腺癌细胞MDA-MB-453的诱导凋亡作用。[方法]免疫组化法检测乳腺癌细胞MDA-MB-453的Her-2表达水平,用流式细胞仪检测细胞凋亡率,比较恩度、紫杉醇单独及联合作用于MDA-MB-453细胞24、48h的细胞凋亡率。[结果]MDA-MB-453细胞Her-2表达率95%可信区间为63.32%~69.60%;恩度与紫杉醇对MDA-MB-453细胞均有诱导凋亡作用,并成剂量依赖关系,其中以恩度50μg/ml联合紫杉醇50nmol/L作用48h组凋亡率最高。[结论]体外环境下恩度、紫杉醇均可诱导Her-2过表达乳腺癌细胞凋亡,且呈剂量依赖关系,两药联合时凋亡作用增强。
[Objective] To observe the effect of Endue and paclitaxel on inducing apoptosis of Her-2 overexpressing breast cancer cell line MDA-MB-453. [Method] The expression of Her-2 in breast cancer cell line MDA-MB-453 was detected by immunohistochemistry. The apoptosis rate of MDA-MB-453 cells was detected by flow cytometry. 24,48h apoptosis rate. [Results] The 95% confidence interval of Her-2 expression in MDA-MB-453 cells was 63.32% -69.60%. Endostar and paclitaxel induced the apoptosis of MDA-MB-453 cells in a dose- Among them, the apoptosis rate was the highest in 48h group with Endostar 50μg / ml combined with paclitaxel 50nmol / L. [Conclusion] Endostar and paclitaxel can both induce the apoptosis of Her-2 overexpressing breast cancer cells in vitro and in a dose-dependent manner.
