采用Northernblot分子杂交技术,检测35例垂体腺瘤和1例正常垂体组织nm23基因mRNA表达。所有垂体腺瘤和正常垂体组织均表达0.8kb的nm23H1mRNA和0.45kb的nm23H2mRNA片段。侵袭型垂体腺瘤nm23H1和H2mRNA表达水平(25.75±17.40,41.58±19.26)分别较非侵袭型垂体腺瘤(46.87±23.51,59.83±25.69)下降,其差别有显著性意义。(P<0.01,P<0.05)。nm23基因mRNA表达水平下降与患者的性别、年龄以及Hardy和Wilson分级、分期无显著相关性。结果提示nm23基因表达与垂体腺瘤的临床行为有关,可作为垂体腺瘤侵袭性敏感度的分子标记
Northern blot hybridization was used to detect mRNA expression of nm23 in 35 pituitary adenomas and 1 normal pituitary. All pituitary adenomas and normal pituitary tissues expressed 0.8 kb of nm23H1 mRNA and 0.45 kb of nm23H2 mRNA fragment. The expression levels of nm23H1 and H2 mRNA in invasive pituitary adenomas (25.75±17.40, 41.58±19.26) were higher than those in noninvasive pituitary adenomas (46.87±23.51, 59.83±25. 69) Drop, the difference is significant. (P<0.01, P<0.05). There was no significant correlation between the decrease in nm23 mRNA expression and the patient’s gender, age, and Hardy and Wilson classification and staging. The results suggest that nm23 gene expression is associated with the clinical behavior of pituitary adenomas and may be used as a molecular marker for the sensitivity of pituitary adenoma invasiveness.
