目的探讨艾迪注射液对顺铂所致大鼠急性肾毒性的保护作用。方法 40只雄性SD大鼠随机分为5组,分别为对照组,顺铂组,艾迪低、中、高剂量组(5、10、15 m L·kg~(-1))。除对照组外,其他组大鼠均于第3天单次腹腔注射给予顺铂10 mg·kg~(-1),艾迪各剂量组分别给予艾迪注射液5、10、15 m L·kg~(-1)连续7 d;正常组和顺铂组给予等体积生理盐水。第8天测定大鼠血清肌酐(Scr)和尿素氮(BUN)含量,检测肾脏组织超氧化物歧化酶(SOD)、谷胱甘肽还原酶(GSH-Px)、丙二醛(MDA)水平,HE染色切片观察肾组织病理变化。结果与对照组相比,顺铂组大鼠血清Scr、BUN和MDA含量明显上升,SOD和GSH-Px含量显著下降,病理损伤严重,肾小管上皮细胞凋亡,并出现炎症;艾迪低、中、高剂量组均能改善大鼠肾脏组织形态,艾迪低剂量组大鼠血清Scr、BUN含量显著下降,SOD和GSH-Px活性显著升高;艾迪中剂量组SOD和GSH-Px活性升高;艾迪高剂量组除了能增强GSH-Px活性外,对其他指标影响均无统计学意义。结论艾迪低、中剂量组对顺铂所致的大鼠肾毒性均有保护作用,其作用机制可能与其抗氧化作用有关。
Objective To investigate the protective effect of Aidi Injection on acute nephrotoxicity induced by cisplatin in rats. Methods Forty male Sprague-Dawley rats were randomly divided into five groups: control group, cisplatin group, low, middle and high dose of Eddie group (5, 10, 15 m L · kg -1). Except for the control group, the other groups were given cisplatin 10 mg · kg -1 by single intraperitoneal injection on the third day, and Eddie injection 5,10,15 m L · kg ~ (-1) for 7 consecutive days; normal group and cisplatin group were given equal volume of normal saline. The levels of serum creatinine (Scr) and blood urea nitrogen (BUN) were measured on the 8th day to detect the levels of superoxide dismutase (SOD), glutathione reductase (GSH-Px) and malondialdehyde (MDA) The pathological changes of kidney were observed by HE staining. Results Compared with the control group, the levels of Scr, BUN and MDA, the contents of SOD and GSH-Px in the cisplatin group were significantly decreased, the pathological damage was serious and the tubular epithelial cells were apoptotic and inflamed. The middle and high dose groups could improve the renal tissue morphology of rats. The contents of Scr and BUN in serum of SD rats in low-dose Aidi group were significantly decreased while the activities of SOD and GSH-Px were significantly increased. The activities of SOD and GSH-Px Increase; Addie high dose group in addition to enhancing GSH-Px activity, the other indicators had no significant effect. Conclusion The low and middle dose of Eddie has the protective effect on nephrotoxicity induced by cisplatin, and its mechanism may be related to the anti-oxidative effect.
