本文报道了用HIV外膜糖蛋白120(gp120)相应区域的同源序列制备合成肽的抗血清,以研究gp120第二稳定区对HIV感染以及抗体中和作用的重要性。首先按照HTLV-Ⅲ_B gp120的氨基酸序列合成三种相重迭的寡肽—T19V、C21E和S19C,并分别与钥孔(虫戚)血蓝素(KLH)结合,免疫兔后制备各自相应的抗血清。所得三种抗血清均能与由感染HTLV-Ⅲ_B的Molt-Ⅲ细胞120或160KD糖蛋白(后者为前者的前身)发生免疫沉淀反应,但不能沉淀未感染的Molt-Ⅲ细胞蛋白,因此看来它们属于gp120和其前身-gp160。
In this paper, antisera of synthetic peptides were prepared using the homologous sequences of the corresponding regions of HIV envelope glycoprotein 120 (gp120) in order to study the importance of gp120 second stable region for HIV infection and antibody neutralization. Firstly, three overlapping oligopeptides-T19V, C21E and S19C were synthesized according to the amino acid sequence of HTLV-Ⅲ_B gp120 and were respectively combined with keyhole limpet hemocyanin (KLH) to prepare their corresponding anti-rabbit serum. All of the three antiserums obtained were immunoprecipitated with 120 or 160 kD glycoprotein from Molt-III cells infected with HTLV-III_B (the latter being the former precursor), but could not precipitate uninfected Molt-III cell proteins They belong to gp120 and its predecessor-gp160.
